While this evidence appears in contrast to the strong association of E4 with risk of Alzheimer's disease (Corder et al., 1993) and to published work on the possible impact of APOE polymorphisms in sCJD showing an overrepresentation of the E4 allele in sCJD patients (Amouyel et al., 1994; Van Everbroeck et al., 2001), such findings have not been replicated in all published studies (Nakagawa et al., 1995; Zerr et al., 1996) nor in our own observations (387 sCJD patients versus unaffected controls, unpublished data).
